Taking peptide recovery ratio as target index, response surface methodology (RSM) was applied to optimize the conditions of enzymatic hydrolysis for Mytilus edulis. According to the principle of quadratic orthogonal rotating combinatorial design, enzyme (E) and substrate (S) mass ratio, enzymatic hydrolysis temperature and enzymatic hydrolysis time were chosen as factors to establish a quadfie regression equation for describing the peptide recovery ratio. By response surface analysis, the optimum conditions for enzymatic hydrolysis was obtained as follows, Enzyme (E) and substrate (S) mass ratio was 3.5:100, enzymatic hydrolysis temperature was 56℃ for 4.2 h. The peptide recovery ratio was up to 35.04% under the optimum condition, which was close to the pedicted value of 36.27%. These results demonstrate the validity of this extraction process.
