The invention relates to the field of nanometer material science and analytical chemistry, in particular to a surface enhanced Raman scattering nano-probe taking polystyrene as a wrapping shell layer, and a preparation method of the surface enhanced Raman scattering nano-probe. The probe comprises an inner layer core, Raman reporter molecules adsorbed on the inner layer core, and the outer wrapping shell layer. The preparation method comprises the following steps of firstly, preparing a noble metal nano-material; then adsorbing the Raman reporter molecules on the surfaces of nano-particles; and finally, by taking the styrene as a monomer, divinylbenzene as a cross-linking agent, and 2, 2-azobis(2-methylpropionamidine)dihydrochloride as an initiator, preparing the polystyrene wrapping shelllayer. The inside of the shell layer is wrapped with the Raman reporter molecules, and the outer polystyrene shell layer separates the reporter molecules from an external environment, so that a signalcannot be disturbed; and the surface enhanced Raman scattering nano-probe has relatively high stability and sensitivity.
